T-bet expression and nuclear mobilization in resting memory CD4⁺ T cells. Memory CD4⁺ T cells were isolated to >95% purity by MACS separation. Cytoplasmic and nuclear fractions from resting memory CD4⁺ T cells were isolated according to the manufacturer's instructions. Memory CD4⁺ T cells were activated with or without immobilized anti-CD3 and anti-CD28 for 12 h, and the levels of IFN-γ and GAPDH mRNA were determined by PCR (a). The ratio of T-bet to GAPDH was quantified by densitometry (b). Means are shown for the results from three independent experiments.
