Time-resolved 1H NMR analysis of Pyl reaction showing the conversion of unbound 2OG (i.e. unreacted substrate) to UDP-Yelosamine-bound 2OG (i.e. 2OG-ligated to the 4-amino-UDP-sugar). Following Pat reaction (time 0), Pyl was added with ATP, MgCl2, and 2-oxoglutarate, and reaction was carried out for 10 min at 25 °C. A chemical shift change of H3 and H4 of 2OG is shown due to attachment to the 4-amino group of the FucNAc-4N of UDP-Yelosamine.
