Library construction. (A) The size of the library was determined by counting the number of clones after gradient dilution and the library size was 5 x 10⁸ cfu/mL. (B) 24 colonies were randomly selected to estimate the correct insertion rate by performing PCR. The size of products was approximately 400 bp.
