A site-directed mutagenesis was made in HMG at position 718 (T718→A718, mHMG) and (A) periodic acid–Schiff staining was performed to detect the galactosylated modifications of HMG. (B) R-250 staining. Lane 1: positive control (horseradish peroxidase); Lane 2: negative control (soybean trypsin inhibitor); Lanes 3 and 5: 5 µg and 20 µg of mHMG, respectively; Lanes 4 and 6: 5 µg and 20 µg of HMG, respectively. mHMG, mutated HMG that T718 in the mG-CSF moiety was mutated to A718.
