DEX treatment causes the translocation of NFATc1 to the nucleus. HTM27 cells were plated onto coverslips then treated with ionomycin for 1 h or DEX or EtOH for 15 min, 30 min, 1 h or 2 h. Cells were fixed with methanol then incubated overnight with anti-NFATc1 antibody followed by Alexa 546 conjugated secondary antibody. (A) No treatment, (B) 1 h Ionomycin, (C) 15 min EtOH, (D) 15 min DEX. Arrows indicate cells positive for nuclear labeling of NFATc1. Scale bar = 20 μm. (E) The number of cells with NFATc1 nuclear labeling was counted from 15 different fields of view for each treatment group and the percentage of total cells was determined. n = 3 biological replicates.
