Core requires ATGL to inhibit LD-associated hydrolase activity from murine liver. A, schematic of the experimental procedure in mice. B–D, WT and ATGL−/− mice were injected with adenovirus expressing core and GFP or GFP alone. Livers were harvested 4 days after infection. B, TG content of liver lysates was measured and normalized to total liver protein (n = 4–5 mice/group). C, LDs were isolated using density gradient centrifugation. Equal amounts of TG were incubated for 1 h at 37 °C. Thereafter, samples were adjusted to 1% Triton X-100 and centrifuged for 30 min at 20,000 × g and 4 °C. FFA in the underlying solution was determined with commercial kits (n = 4). Data are presented as the mean ± S.D. D, the presence of GFP and core in the cytosolic and LD fractions, respectively, was confirmed by Western blot.
