Transient transfection of shRNA-Ascl2/LS174T cells with miR-200 family member inhibitors increased the expression of mesenchymal markers or reduced the expression of epithelial markers as indicated by quantitative real time PCR. The mRNA levels of E-cadherin, N-cadherin, snail, slug, ZEB1, and ZEB2 in shRNA-Ascl2/LS174T cells transfected with the miR-200b inhibitor (A), miR-200a inhibitor (B), miR-429 inhibitor (C), miR-200c inhibitor (D), miR-141 inhibitor (E), or negative control (NC) inhibitors and untransfected shRNA-Ascl2/LS174T cells (Con) were determined via real time PCR in three independent experiments. β-Actin was used as the control (*, p < 0.05; **, p < 0.01).
