Impact of double deficiency of Src family kinases (SFK) in glycoprotein VI (GPVI)-induced platelet aggregation. (A) Washed platelets (2 × 108 mL−1) prepared from fyn−/−lyn−/− (top), fgr−/−lyn−/− (top), fyn−/−src−/− (middle) or lyn−/−src−/− (bottom) mice and their respective litter-matched control were stimulated with different doses of CRP (0.3, 1 and 3 μg mL−1). Platelet aggregation was measured as a change in light transmission and ATP secretion was measured as luciferin/luciferase-mediated luminescence, using a lumi-aggregometer. Representative images are shown (n = 4 mice per condition). (B) Time to 25% of aggregation was calculated and data are shown as mean ± SEM.
