InsP₃R3–3′-UTR-Luciferase reporter activity is inhibited by miR-506 mimics (A) but not by miR-124 in HEK293 cells (B). A, HEK293 cells were transiently transfected with InsP₃R3–3′-UTR-Luc or a control vector and co-transfected with 50 nm miR-506 mimic or mimic and inhibitor. Luciferase activity after 48 h was assayed as described under “Experimental Procedures.” miR-506 mimic inhibited reporter activity, which was abrogated by inhibitor coexpression. B, under the same conditions as in A, miR-124 mimic or mimic + inhibitor expression had no specific effects on the reporter activity suggesting a specific effect of miR-506 on InsP₃R3 mRNA.
