The central role of MSX2 in ameloblast cell fate. Msx2 expression is controlled by transcription factors involved in ameloblast proliferation. MSX2 modulates the expression of its target genes directly or indirectly by interacting with various partners such as C/EBPα or Dlx (Zhou and Snead, 2000; Lézot et al., 2002, 2008). Thus, it represses the transcriptional activity of the transcription factors that modulate amelogenin expression.
