Effects of celastrol treatment on lipolysis and intracellular lipid accumulation of differentiated 3T3-L1 adipocytes with mitochondrial dysfunction. In the presence of MDI, fully differentiated 3T3-L1 adipocytes were incubated with oligomycin and celastrol for 48 h differentiated cells were on analyzed for (A) free fatty acids and (B) glycerol release as described in materials and methods. The Oil Red O stained adipocytes were photographed at 40× magnification; (C) Untreated control (DMSO); (D) MDI-treated cells; (E) Oligomycin-treated cells and (F) Oligomycin + celastrol-treated cells. Figures represent one of three independent experiments; and (G) Quantification of intracellular lipid droplets in 3T3-L1 adipocytes treated with oligomycin and celastrol was performed accordingly. The results are representative of those obtained from three independent experiments.
