CCP1 overexpression increases the levels of Δ2-tubulin. A, CCP1 transfection in HEK293T cells. Protein extracts from control cells or cells expressing HA-tagged CCP1 were prepared. Samples (30 μg of protein extract) were analyzed by Western blot using an anti-HA antibody. Besides the main 150-kDa band, additional bands of ∼120–130 kDa and 80–90 kDa can be observed, similar to the patterns previously reported in literature (2, 37). B, Detection of tubulin PTMs. Extracts from control cells or CCP1 expressing cells were analyzed by Western blot using anti-Δ2-tubulin and anti-Tyr-tubulin antibodies. The Western blot probing anti-β-actin served as loading control.
