Effects of EPA and TUG-891 on migration of DU145 (A) and PC-3 (B) cells. Serum-starved DU145 and PC-3 cells were seeded in serum-free medium at 2.5 × 104 cells per insert in the upper chambers of 8-μm transwell inserts. Serum-free medium was added to the lower wells. Cells were then treated with 10% FBS, 20 μM EPA, 1 μM TUG-891, 10 μM LPA, or 10 nM EGF, either alone or in combination. Following a 6-hour migration period, the insert membranes were fixed and stained using methanol and crystal violet. Cells that invaded the lower chambers were counted by microscopy. Each bar represents the mean ± S.E.M. of values obtained from duplicate experiments, each of which used duplicate wells of cells for each experimental condition. Data analysis was performed using one-way ANOVA, followed by Tukey’s multiple comparisons test.
