Compound 4 inhibits iTreg generation. (A) Naïve wild-type CD4+ T-cells were activated for three days (Dynabeads plus iTreg polarizing cytokines, IL-2 + TGF-β) by increasing concentrations of 4 (CVN). Agonist OX40 antibody (OX86) and tartrazine (TZ) were added as positive and negative controls respectively. Inhibitory effect on the induction of iTreg cells were accessed by gating on the live CD4+ T-cell population. (B) Summary of all experiments showing average ± SD of three independent replicates. Data were analysed by anova with Dunnett's post hoc test with *P < 0.05 and **P < 0.01 as compared with Treg polarizing conditions (IL-2 + TGF-β).
