Role of the C-terminal β-hairpin of R. sphaeroides RbcS in Rubisco stability and assembly. A, thermal stability of Rubisco containing WT RsRbcS or RsRbcSΔEF. RbcL and RbcS were co-expressed in E. coli, and lysates were incubated at the indicated temperatures for 10 min, followed by cooling on ice and activity assay at 25 °C. Activities measured after incubation at 30 °C were set to 100%. Shown is S.D. from at least three independent experiments. B, carboxylation activity during in vitro reconstitution of RsRubisco with WT RsRbcS or RsRbcSΔEF. Experiments were performed essentially described as in Fig. 2C (panel iii).
