Quantitatively normal response to ECTV by unhelped secondary anti-VACV CD8M maintained in the absence of help. B6 mice and MHC-II0/0 mice were primed and boosted 5 weeks later with 5 × 106 PFU VACV i.v. At 60 dpp or dpb, the splenocytes were obtained and labeled with CFSE, and the CD8+ T cells were magnetically purified, equalized for similar numbers of Kb-TSYKFESV+ CD8+ T cells, and inoculated i.v. into B6.Thy1.1 mice. One day later, the mice were infected with 3 × 103 PFU ECTV in the footpad or left uninfected as a control. Cells in the draining popliteal lymph node were analyzed at 5 days p.i.. Representative flow cytometry plots on the left show the frequencies of transferred cells within the CD8+ gate (top row), the frequencies of cells within the transferred cells that diluted their CFSE (middle row), and the frequencies of cells that diluted CFSE and express GzB and IFN-γ.
