Immunocapture of NL4.3-derived WT and mutant viruses produced in primary human cells. NL4.3-derived WT virions and mutant virus particles carrying the listed alanine substitutions were obtained from mitogen-treated PBMCs from three different healthy donors. Virus infection was allowed to proceed for 5 days before capture of progeny virus with magnetic beads coated with either an anti-ICAM-1 antibody (R6.5) or an isotype-matched irrelevant biotinylated antibody (used as a control). Precipitated viral entities were quantified with a homemade p24 ELISA. Data shown represent the means ± standard deviations of data from triplicate samples and are representative of three independent experiments. The percentage of ICAM-1 incorporation for each individual donor is denoted on each corresponding graphic.
