Mutation of MALT1 at K644 decreases its activity. HEK-293A cells were transfected as indicated and lysed in the presence of Cy5-LVSR-AOMK, followed by SDS-PAGE of total cell lysates (no separation into NP40-soluble and insoluble fractions), fluorescence scanning, and Western blot analysis with the indicated antibodies. Note that the antibodies detect both endogenous and Flag-tagged versions of the proteins, which migrate at a slightly different MW. In addition, the MALT1 antibody detects MALT1 cleavage products of unknown significance and the Bcl10 antibody detects multiple higher MW species, which most likely represent posttranslationally modified variants.
