Blood glucose is increased in GLS5 mice fed a LD. Male mice (10 weeks old) were fed a LD for 10 days. A, C: Blood was collected retro-orbitally from 4 h-fasted mice (n = 4–6/group). A: Glucose was determined using an enzymatic colorimetric assay. B: Relative hepatic mRNA levels of gluconeogenic genes Pck1 and G6pc were measured. C: Plasma insulin levels were measured using ELISA in samples collected as described for (A). D: Glucose tolerance tests were performed in 4 h-fasted male mice. Mice were intraperitoneally injected with 2 g/kg body weight glucose and blood glucose was measured using a glucometer at 0, 30, 60, 90, and 120 min postinjection. Values are mean ± SEM. *P < 0.05 versus WT, #P < 0.05 versus GKO; one-way ANOVA analysis with Tukey’s post hoc test.
