E14 mesencephalic cultures were treated with 5 nM rotenone or vehicle control on DIV 6. After 12 h, live cells were stained with CM-DCFDA to monitor ROS levels, photographed, then fixed and immunostained for TH for analysis. Images of CM-DCFDA and TH staining were superimposed. Cells stained positive for both were identified; the relative intensity of CM-DCFDA staining in TH+ cells was quantified.
