Comparison of anti-inflammatory activities of ANO and AR-selective agonists. Peritoneal macrophages (5 × 104/well) were stimulated with LPS (1 μg/mL) and muIFN-γ (10 IU/mL) in the presence or absence of various concentrations of ANO or A1 AR-, A2A AR- and A3 AR-selective agonists (CCPA, CGS21680 and IB-MECA, respectively) at 37°C for 24 h (A). Sodium butyrate-treated THP-1 cells (1 × 105 /well) were stimulated with LPS (5 μg/mL) plus huIFN-γ (500 IU/mL) in the presence or absence of various concentrations of ANO or AR-selective agonists at 37°C for 24 h (B). Levels of TNF-α, IL-6 and IL-12 in the culture supernatants were determined by ELISA. Values represent the percentage of control culture. Results are representative of two separate experiments with similar results.
