Rescue of plasmid-based reverse genetics system for ISAV. Shown is the detection of segment 6 with NotI from supernatants of ASK cells cotransfected with 8 pSS-URG genomic segment (segments 1 to 8) plasmids with (8pSS-URG + 4P) or without (8pSS-URG) four plasmids expressing the proteins of RNPC. The supernatant from transfected (P0) and infected cells after the first two blind passages with the rISAV were analyzed by RT-PCR 7 days postinoculation. Data show triplicate transfections (I to III) for each condition. Products were visualized by agarose electrophoresis.
