Chimera analysis reveals that WAVE1 inhibitory effects on filament elongation stem from its WH2 (V) domain. (A) TIRF analysis of mean elongation rates for filaments polymerized in the presence of GST-VCA chimeras between bovine N-WASP and human WAVE1. Reactions contained 1 μM G-actin (10% Oregon green labeled), 3 μM profilin, and 0 or 100 nM GST-VCA. From 11 to 15 filaments per reaction were analyzed for each of three independent trials.
