Nfasc140 rescues the PNS and CNS nodal complexes as efficiently as Nfasc186 in Nfasc−/− mice. Immunostaining of teased quadriceps (PNS) and spinal cord ventral funiculus fibers (CNS) at P60 shows that both FLAG-tagged Nfasc140 and Nfasc186 can rescue the nodal components, voltage-gated sodium channels (Nav), ankyrinG, and βIV-spectrin when expressed on a neurofascin-null background. Additionally, the PNS-specific nodal proteins NrCAM and gliomedin were also rescued, as was the CNS-specific nodal protein brevican. Rescue of the node was not accompanied by reformation of the axoglial junction; and, as a consequence, juxtaparanodal Kv1.1 channels were mislocalized compared with WT. Scale bars, 5 μm.
