Quantification of cleaved 8-OH-Gua. After in vitro oxidation with 10 μM Fe(II)/ascorbate or 10 μM cyt c and the indicated concentrations of H2O2, oxidized RNA was precipitated with ethanol. The supernatant was mixed with internal standards and then trimethylsilylated without acid hydrolysis. The resulting mixture was analyzed with GC/MS. The quantities of 8-OH-Gua generated are presented as percentage increase relative to that observed with control. The results represent means ± SD in triplicate experiments. *p < 0.05 relative to the nonoxidized control.
