Increased viral replication results from mutating miR-U86. To assess viral replication, sorbitol-purified virus (WT, ΔmiR-U86, and ΔmiR-U86 REV) was used to infect Jjhan cells (MOI = 0.01). Infected cells were harvested for qPCR at 3, 6, and 9 days p.i., and viral genomes were detected using primers directed at GFP and normalized to cellular MDM2. All values are shown relative to input.
