Schematic of the detection and characterization study of CTC and metastatic tumor using magnetic nanoparticle labeling and quad-µNMR detection system. Peripheral blood (7 ml) and fine needle biopsy from the site of metastasis were obtained. Samples were labeled by incubating cells with primary antibodies followed by TCO-modified secondary antibodies (Step 1). Cells were then pelleted by centrifugation and resuspended directly with Tz-nanoparticles that selectively targeted TCO-antibodies (Step 2). The process of labeling and targeting required 40 minutes. Cancer marker measurements were taken using the µNMR device to produce quantitative protein readouts.
