YM-53601 does not affect NS4B foci in Huh-7.5.1-8 cells. Huh-7.5.1-8 cells that were stably transfected with modified pCXN2 (A and B) or pCXN2/HA-TEV-NS4B (C and D) were grown on coverslips and treated with 1.5 μM YM-53601 (B and D) or DMSO (A and C) in serum-free medium for 3 days. The cells were fixed and subjected to immunofluorescence analysis using confocal microscopy. HA-tagged NS4B protein was detected with a rat anti-HA antibody followed by an Alexa Fluor 488-conjugated anti-rat antibody (green), and the nucleus was stained with DAPI (blue).
