Direct detection of base J through SMRT DNA sequencing. Synthetic oligonucleotides containing J at known positions were subjected to SMRT sequencing, and the associated polymerase kinetics were compared with those obtained with a template of identical sequence but lacking the base J modification (and containing T at that position). The direction of DNA polymerase is indicated. The Y-axis shows the fold change in polymerase kinetics (interpulse duration (IPD) ratio) at each position between modified and unmodified sequences.
