(a,b) WT bone marrow-derived macrophages were pre-incubated with cholesterol (20 μg/ml) or 7-KC (5 μM) for 2 hours. After that, the cells were stimulated with IFNγ (20 ng/ml) and LPS (100 ng/ml) or left unstimulated as described in the Methods. MHC II expression was analyzed by flow cytometry and TNFα production was analyzed by ELISA. (c) Representative plot and (d) graph show levels of NF-κB p65 phosphorylation (Ser 529) in Abcg1−/− and WT BMDMs analyzed by flow cytometry.
