Systematic comparisons of the transfection efficiencies between modified and traditional procedures. (A) Quantitative analysis of the transfection efficiency in different cell strains. Plate: six-well format. Complex: polyethyleneimine (PEI)/pEGFP-C1. Modified: 700 μL of polyplex was added to cells without medium. Traditional: 500 μL polyplex was added to cells cultured in 200 μL fresh medium. Transfection efficiencies were analyzed by fluorescence-activated cell sorting 48 hours after transfection. (B) and (C) qualitative evaluation of the transfection efficiencies in different complexation volumes using Lipofectamine™ 2000/pEGFP-C1 lipoplexes (B) and PEI/pEGFP-C1 polyplexes (C). Cell line: Hep G2. Plate: 24-well format. Magnification: 100×. Time: 48 hours after transfection. Modified: the complexation complexes with indicated volumes were added to cells without medium and incubated for 1.5 hours. Traditional: 100 μL complex was added to cells with medium to get indicated final volumes from 200 μL to 500 μL.
