FIG. 1.

FIG. 1.

NF-κB decoy ODN treatment increased mouse MSC viability but not cell apoptosis in response to UHMWPE particles. Mouse MSCs were exposed to (a) control or (b) 1 μg/mL LPS, together with 0.5 μM decoy ODN or scrambled ODN, with or without UHMWPE particles. Cell viability was quantified at 48 h using the LDH assay. (c) Mouse MSCs exposed to ODN, UHMWPE particles, and LPS for 24 h were stained with Annexin V to evaluate the cellular apoptotic response. The apoptotic cells were analyzed by flow cytometry. The error bars represent the SEM. *p<0.05, **p<0.01, and ***p<0.005. LDH, lactate dehydrogenase; LPS, lipopolysaccharide; MSCs, mesenchymal stem cells; ODN, oligodeoxynucleotide; SEM, standard error of mean; S-ODN, scrambled ODN; UHMWPE, ultrahigh-molecular-weight polyethylene; UNT, untreated control.
