Pharmacologic inhibition of the Notch signaling pathway using the γ-secretase inhibitor (GSI) MRK003. (A) HES1 in cell lines Res186 and Res259 is inhibited at the mRNA level after 48-hour treatment with GSI MRK003. (B, C) Scratch test assays demonstrate that Res186 migration is significantly inhibited after 17 hours at 5 µM GSI MRK003, although Res259 trends slightly slower, it is not statistically significant. All experiments were performed in triplicate; cells were treated with GSI MRK003 in 2% fetal bovine serum (*p < 0.05, **p < 0.005, ***p < 0.0005).
