Fig 2 — Ras61p with several CaaX-box motives known to be farnesylated, geranylgeranylated or both were analysed for their ability to complement growth in the RRS. Proteins known to be farnesylated generally rescued growth while the unprenylatable motif-SRSA did not. This includes the mono-geranylgeranylated motives-KCAIL of CDC42p and -TCTIL of Rsr1p which are known to be cross-farnesylated, but not-KCVLL of Rho1p which is exclusively geranylgeranylated. This suggests that only farnesylation is detected in the RRS (+Met denotes 5 μg/mL methionine in the medium to suppress gene expression while in its absence gene expression is induced).
