PKG Iβ binds exclusively to HAP1 harboring the predicted anchoring domain. Western blot analysis of lysates and proteins pulled down with the 8-AHA-linked cAMP-agarose beads in COS-7 cells co-expressing PKG Iβ-GFP plasmid with (i) the HAP1-DSRed construct (Control), (ii) the HAP1-DSRed bearing the double mutation K567A/R568A, and (iii) the C-terminal truncated HAP1-DSRed construct (HAP1 amino acids 157–386) is shown. Pulldowns also were performed in non-transfected cells and cells transfected with only PKG Iβ or HAP1 as negative controls.
