Human light and heavy chain pseudogene sequences. (A) Alignment of SynVK pseudogenes in the SynVK-C construct. Top line shows the sequence of the functional HuVK gene that is mutated by the SynVK pseudogenes. The CDRs (boxes; IMGT nomenclature) were derived from human EST databases. Some pseudogenes also contain framework changes derived from the ESTs. At the 3′ end of the pseudogenes, the sequence of the flanking DNA downstream of CDR3 in each pseudogene is shown. This flanking sequence is part of the 100 bp spacer sequence inserted between each pair of pseudogenes. (B) Alignment of the SynVH pseudogenes in the SynVH-B construct. Top line shows the sequence of the functional HuVH gene. The CDRs consist of a tyrosine/tryptophan/serine scan. The framework regions contain no changes.
