Interleukin-21 (IL-21) decreased cytokine production of polarized T helper type 2 (Th2) cells. Polarized Th2 cells (5 × 105 cells) were re-stimulated with ovalbumin 323–339 (OVA323–339) peptides pulsed splenocytes in the presence of IL-21 or IL-21 plus IL-21R:Fc. (a) Th2 cytokines IL-4, IL-5 and IL-13 in the supernatants were detected by ELISA. (b) GATA3 expression in Th2 cells was measured by quantitative RT-PCR. (c) Apoptosis of Th2 cells was measured by staining with annexin V and 7-aminoactinomycin D (7-AAD) and analysed with a flow cytometer. The apoptotic cells were annexin V+ 7-AAD and annexin V+ 7-AAD+. (d) Bcl-2 expression of Th2 cells was measured by quantitative RT-PCR.
