Residue-specific and site-specific labeling of Mel-SH527-IIA4 with BODIPY-TMR-lysine. Cell-free synthesis was performed in presence of ¹⁴C-leucine using the insect cell-free expression system. De novo synthesized scFv antibody fragments were visualized by in-gel fluorescence (extinction 532 nm, emission filter 580 nm bandpass, Typhoon TRIO+ Imager, GE Healthcare) and autoradiography.
