(i) Yeast Atg4 is a cysteine protease required for the activation of immature Atg8 protein, cleaving its C-terminal region and exposing a glycine residue (GLY116) that is essential for its attachment to the phagophore. (ii) Atg4 is also responsible for the recovery of Atg8 via delipidation of this protein from mature autophagosomes or organelle membranes, where it can be erroneously attached.
