Model of WISp39 function. Unphosphorylated Coronin 1B binds Arp2/3 complex (step 1; Cai et al., 2005) and localizes it to the side of the actin filament (step 2) but prevents branching by Arp2/3 complex (Humphries et al., 2002). Coronin 1B phosphorylated by PKC on Ser2 loses its affinity for Arp2/3 complex (Cai et al., 2005; this paper), allowing Arp2/3 complex-mediated branching (step 3). Phosphorylated Coronin 1B binds WISp39 in a complex with SSH and Hsp90 (step 4; this paper). SSH dephosphorylates Coronin 1B and Cofilin (step 5; Cai et al., 2007; this paper). Dephosphorylated Coronin 1B binds Arp2/3 complex and removes it from the branch (step 6; Cai et al., 2008). The destabilized branch is then severed by active Cofilin (Pollard and Borisy, 2003).
