FIG 3

Localization of PyExp2 containing a C-terminal HA tag is unaltered in P. yoelii 17X-infected RBCs. (A) Immunoblot analysis of P. yoelii 17X parasite lysate and transgenic P. yoelii 17X-Exp2-3HA parasite lysate probed with rabbit anti-rPyExp2 sera, mouse anti-HA monoclonal antibody, or rabbit anti-rPyMSP8 sera. Parasites obtained by saponin lysis of infected RBCs were solubilized in SDS-PAGE sample buffer, and equal volumes of parasite lysate were loaded on the gel. Molecular mass markers are indicated. (B) Immunofluorescence images showing localization of PyExp2-3HA fusion protein (green) in RBCs infected with transgenic P. yoelii 17X-Exp2-3HA parasites using mouse anti-HA monoclonal antibody. DAPI was used to stain the parasite nuclei (blue).
