Ten nM pro-uPA was incubated with 100 nM mAb-PUK (black triangles), 100 nM anti-PAI-1 mAb-2 (white circles) or without antibody (black circles). At time zero, 0.5 nM plasmin or 5 nM matriptase was added, followed by incubation of the mixtures at room temperature for the indicated time periods. Finally, the activity of plasmin or matriptase was quenched by the addition of aprotinin to a final concentration of 1 µM. S-2444 was added to a final concentration of 0.5 mM and the amount of active uPA formed was estimated by the rate of hydrolysis and comparison to a standard curve of active two-chain uPA.
