Hyperglycemia and acute glycemic fluctuation enhances MG occludin glycation. IHECs were grown under normal glucose (5 mM) or high glucose (25 mM glucose for 7 days or 12 days) or in cells subjected to acute glycemic fluctuation (25 mM glucose for 12 days, then change from 25→5 mM glucose for 6 h). Total occludin and MG–occludin glycated adducts were determined in whole cell lysates by Western blot analyses. Representative blots were shown.
