Inhibition of Hsp90 or IKK-i impairs Sendai virus-induced apoptosis. (A and B) HEK293 cells were treated with DMSO, RA (2.5 μM) (A), or GA (2 μM) (B) and then infected with SeV (left) or treated with TNF-α (10 ng/ml) and CHX (10 ng/ml) (right) for the indicated times. Cell lysates were collected and subjected to immunoblot analysis with the indicated antibodies. (C) HEK293 cells were transfected with NC or Hsp90 siRNA and then infected with SeV (left) or treated with TNF-α (10 ng/ml) and CHX (10 ng/ml) (right) for the indicated times. Cell lysates were collected for Western blot analysis with the indicated antibodies. (D) HEK293 cells were treated with BX795 (2.5 μM) or DMSO and infected with SeV (left) or treated with TNF-α (10 ng/ml) plus CHX (10 ng/ml) (right) for the indicated times. Cell lysates were collected and subjected to immunoblot analysis with the indicated antibodies. (E) HEK293 cells were transfected with the indicated siRNAs and then infected with SeV (left) or treated with TNF-α (10 ng/ml) and CHX (10 ng/ml) (right) for the indicated times. Cell lysates were collected for Western blot analysis with the indicated antibodies. NC, negative-control siRNA; *, nonspecific band.
