Direct regulation of HOXA11 by miR-30d and 181a. 293T cells were transfected with luciferase reporter plasmid containing 3′-UTR of HOXA11, together with miRNA mimic, anti-miRNA or negative controls. MiR-30d or 181a overexpression significantly decreased the relative luciferase activity of the wild-type 3′-UTR but not the mutant 3′-UTR in which these two miRNA-binding sites were deleted (A). Conversely, knockdown of miR-30d or 181a significantly increased the relative luciferase activity (B). Each experiment was conducted in triplicate.
