Activation of human leucocyte antigen (HLA)-A2-restricted NY-ESO-1-specific T cell receptor (TCR)-transduced CD8⁺ T cells by peptide-pulsed target cells. HLA-A2⁺ CD8⁺ T cells transduced with the indicated NY-ESO-1-specific TCRs were stimulated with HLA-A2⁺ C1R target cells pulsed with the cognate SLLMWITQC peptide across a range of concentrations. Four functional readouts [macrophage inflammatory protein (MIP)-1β, tumour necrosis factor (TNF)-α, interferon (IFN)-γ and interleukin (IL)-2] were measured by flow cytometry. Pie chart segments represent the fraction of cells expressing the number of functions indicated in the key. The gating strategy followed that described for Fig. 1a, with the exception that T cell antigen receptor variable (TCRV)β13·1 expression was used to identify transduced T cells where appropriate.
