(A) Whole-embryo irradiation of zebrafish mounted in an agarose template using a Leica DM4500B upright fluorescence microscope equipped with a 20x, 0.5 NA water-immersion objective. (B) An embryo injected with mRNA encoding the photoconvertible protein Kaede and globally irradiated from the animal pole for 10 sec at 7 hpf. Fluorescence imaging revealed more Kaede photoconversion at the animal pole then at the margin. (C) The same embryo globally irradiated laterally, as mounted in (B), for an additional 10 sec, resulting in even distribution of photoconverted Kaede protein. Scale bar: 200 μm.
