PP2APpp2r2d protein expression is responsive to cholesterol level. A, A7r5 cells were incubated with MCD for the indicated times, and the cholesterol level was determined using a cholesterol assay kit. B, A7r5 cells that were treated with MCD for 2 h were incubated with cholesterol-loaded MCD (MCD-CHOL) for the indicated times, and the cholesterol level was determined using a cholesterol assay kit. C, the protein expression of various PP2A subunits, pAMPKα, AMP kinase, and Acc1 (pAcc1) phosphorylation was determined in A7r5 cells at the indicated times using Western analysis. The antibodies used were specific for each subunit, and they were tested for any cross-reactivity using siRNA knockdown experiments. Actin was used as a loading control. Error bars represent S.E.
