Positional distribution of [¹⁴C]acetate incorporation in the presence of endogenous fatty acids.

Wild-type strain TIGR4 and MDJ01 (ΔplsX) were grown without or with different fatty acid supplements and labeled with [¹⁴C]acetate (5 μCi/ml) for 3 hours in mid-log phase in CY/BSA medium. Lipids were extracted and PtdGro isolated by thin-layer chromatography and subjected to digestion with Naja naja phospholipase A₂. The digested samples were separated on Silica Gel H thin-layers developed with chloroform/methanol/acetic acid (55/20/5, v/v/v). The distribution of label between the 1- and 2-positions was determined using a Bioscan Imaging detector. The lyso-PtdGro (1-position) peak and fatty acid (FA, 2-position) peaks are labeled.
