Motor performance of L66 mice treated with MTC and LMTX as a function of brain MT concentration. The performance, as measured by the mean time of 12 post-treatment trials spent on RotaRod, for L66 mice treated with 5 or 15 mg/kg MTC (○) or LMTX (●) is plotted as a function of brain MT concentration. Performance and brain concentrations of MT are similar, whether delivered as MTC or LMTX. There is nonlinear improvement in performance over a 10-fold MT concentration range, with evidence of plateauing at MT concentrations of 0.6 mmol/l (MTC) and 1.4 mmol/l (LMTX, in this case LMTB). The 5 and 15 mg/kg doses are equivalent to 3.7 and 11 mg MT/kg for both MTC and LMTB.
